ABSTRACT

【Objective】 To explore the correlation of DAT-positive hemolytic anemia(HA) with gender, age, and ABO serological indicators (anti-A, anti-B, anti-AB, anti-D; Ac, Bc, Oc), so as to provide references for clinical diagnosis. 【Methods】 2 832 HA patients who had underwent DAT tests in the Second Affiliated Hospital of Xi′an Jiaotong University in recent years were divided into the positive-DAT patients ( n=382 )and negative-DAT patients (n=2 450 ) according to the testing results. The correlation between the positive-DAT patients and the sex, age and ABO serological indicators was analyzed using SPSS 25.0 statistical software. 【Results】 DAT-positive HA risk in women were significantly higher than men (OR=1.712, CI95%= [1.365, 2.147], P<0.01), and it differed significantly by ages; it increased significantly with the presence of positive anti-A, negative Ac (OR=1.429, CI95%= [1.150, 1.775] ; P<0.01), and positive Oc(OR=21.369, CI95%= [13.813, 33.057] ; P<0.01). Men aged 31~40 were prone to develop mixed type(OR=19.304, CI95%= [2.208, 168.751] ; P<0.01), women aged 41~50 were prone to cold antibody(OR=3.981, CI95%= [1.545, 10.259] ; P<0.01); anti-AB positive patients were prone to mixed type (OR=1.852, CI95%= [1.027, 3.339] ; P<0.05), Oc positive was prone to warm antibody(OR=4.018, CI95%= [1.386, 1.646], P<0.01). 【Conclusion】 DAT-positive HA is significantly correlated with gender and age, and is positively correlated with anti-A, Oc positive and Ac negative.

ABSTRACT

Background@#Our previous studies have shown that regulatory factor X5 (RFX5), a classical transcription regulator of MHCII genes, was obviously overexpressed in hepatocellular carcinoma (HCC) tumors. However, the role of RFX5 in the carcinogenesis and progress of HCC remains unknown. This study aimed to reveal its biological significance and the underlying mechanism in HCC.@*Methods@#RFX5 mRNA expression level and copy number variation in HCC tumors and cell lines were determined by analyzing deposited data sets in the Cancer Genome Atlas and Gene Expression Omnibus database. The biological significance of RFX5 in HCC was investigated by monitoring the colony formation and subcutaneous tumor growth capacity when RFX5 was silenced with lentiviral short hairpin RNA and CRISPR/Cas9 system in HCC cell lines. The downstream gene transcriptionally activated by RFX5 in HCC cells was determined by chromatin immunoprecipitation and luciferase reporter assay. The involvement of tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein theta (YWHAQ) in HCC development was further determined by performing colony formation rescue assay and subcutaneous tumor growth rescue experiment. The association of YWHAQ with recurrence-free survival of patients with HCC was assessed by Kaplan-Meier analysis. Moreover, apoptosis level and the protein level of p53 pathway were determined to reveal the mechanism of RFX5 in driving HCC development.@*Results@#RFX5 was amplified and highly overexpressed in HCC tumor tissues compared with the corresponding non-tumor tissues. The mRNA expression level of RFX5 was significantly correlated with its DNA copy number (r = 0.4, P < 0.001). Functional study demonstrated that RFX5 was required for both clonogenic forming in vitro and subcutaneous tumor growth in vivo of HCC cells. Further study identified YWHAQ, namely 14-3-3 tau, as a key downstream transcriptional target gene of RFX5, which was tightly regulated by RFX5 in HCC. Moreover, overexpression of YWHAQ largely rescued the clonogenic growth of HCC cells that was suppressed by RFX5 knockdown. In addition, overexpression of YWHAQ in primary tumor was linked to poor prognosis of patients with HCC. These results demonstrated that YWHAQ was a downstream effector of RFX5 in HCC. Notably, RFX5-YWHAQ pathway could protect cells from apoptosis by suppressing the p53 and Bax in HCC.@*Conclusion@#RFX5 is a putative HCC driver gene that plays an important role in the development and progression of HCC by transactivating YWHAQ and suppressing apoptosis.

ABSTRACT

Objective:To explore the effects of suppress or enhancer of lin-12-like(Sel1L) on differentiation and function of bone marrow-derived dendritic cells.Methods:To generate conditional knockout mice by the Cre-Loxp recombination system.ELISA and Real-time fluorescence quantitative PCR(RT-PCR) was used for analyzing the protein levels and mRNA levels of IL-6/IL-12 in BMDCs.The protein levels of Sel1L in BMDCs were detected by Western bolt.The expression of CFSE,CD80,CD86,MHC-Ⅰ,MHC-Ⅱon BMDCs and the capability in priming OVA specific CD4+T cells proliferation were analyzed by the flow cytometry.Results:The deficiency of Sel1L decreases the proliferation of DCs during its differentiation,up-regulates the secretion of IL-6,IL-12 and the expression of MHC-Ⅰ.Notably,Sel1L-null DCs was failed to up-regulate MHC-Ⅱexpression and dramatically impaired their ability to prime OVA323-339specific CD4+T cell.Conclusion:The deletion of Sel1L can reduce the proliferation of BMDCs and down-regulate its ability in priming the proliferation of OVA specific CD4+T cells.

ABSTRACT

Objective To explore the effect of galangin on the inhibition of proliferation and invasion in human lung cancer A549 cell lines and its mechanism thereof. Methods MTS assay was employed to detect the ability of cell proliferation with different concentrations (0, 5, 10, 20, 40, 60 and 100μmol/L) of galangin after A549 cells were cultured in 96 well-plate. Transwell assay was employed to detect the ability of cell invasion. Western blot assay was employed to detect the protein expression levels of Survivin, p27, CD44, ICAM, MMP-2/9 and the phosphorylation of PI3K and AKT. Results The cell proliferation rate of A549 cell line was suppressed with increased concentration of galangin treatment, and IC50 was 44.7μmol/L. The concentrations of 10μmol/L and 20μmol/L were used for the following study. The ability of cell invasion was decreased with 0, 10 and 20μmol/L concentrations of galangin treatment (P<0.05). The protein expression of p27 was increased and the expressions of other proteins were decreased with 0, 10 and 20μmol/L galangin treatmenmt (P<0.05). Conclusion Galangin can significantly inhibit the cell proliferation and invasion of human lung cancer A549 cell line, and which is a potential anti-lung cancer drug. The mechanisms may be related with the regulated related gene expression and the inhibited phosphorylation of PI3K and AKT.

ABSTRACT

BACKGROUND:Tumor necrosis factor-α(TNF-α), a main cytokine inducing chondrocyte apoptosis of osteoarthritis, plays a regulatory role in the process of osteoarthritis. OBJECTIVE:To compare the rat models of chondrocyte apoptosis induced by different mass concentrations of TNF-α, thus providing theoretical basis for further study on the regulation of drugs on chondrocyte apoptosis. METHODS:Chondrocytes were isolated from the knee cartilage of 4-week-old Sprague-Dawley rats of clean grade by mechanical l col agenase digestion and were then incubated with different mass concentrations of TNF-αto induce apoptosis. The morphology of chondrocytes was observed under inverted phase contrast microscope, cel s were identified using immunohistochemical staining of type II col agen, as wel as the cel viability and apoptosis were detected by MTT and DAPI, respectively. RESULTS AND CONCLUSION:(1) In vitro, the cytoplasm of chondrocytes was stained brown-yel ow by using immunohistochemical staining of type II col agen. (2) At 48 hours, the apoptosis rate of chondrocytes induced by 10, 20 and 30μg/L TNF-αwas significantly higher than that of the 0μg/L TNF-α(P<0.01), and the apoptosis rate of chondrocytes induced by 40μg/L TNF-αwas significantly higher than that of the 10μg/L TNF-α(P<0.01). (3) The viability of chondrocytes induced by 10, 20 and 40μg/L TNF-αwas significantly lower than that of the 0μg/L TNF-α(P<0.01). In detail, the viability of chondrocytes induced by 20μg/L TNF-αwas lower than that of the 10μg/L TNF-α(P<0.05);the viability of chondrocytes induced by 40μg/L TNF-αwas significantly lower than that of the 10 and 20μg/L TNF-α(P<0.01, P<0.05). (4) These results suggest that 20μg/L TNF-αcan successful y induce the chondrocyte apoptosis model.

ABSTRACT

Objective To understand the current situation of scientific research integrity level of medical graduate students,the development of integrity education in their universities,to put forward corresponding suggestions for improvement.Methods From December 2015 to March 2016,128 postgraduates are selected as the participants of the survey by randomized method (123 responded).We conducted a questionnaire survey in four famous medical universities in Beijing,descriptive analytical method was adopted to analyze survey findings.The contents of survey included the status of academic integrity of the current medical students,education of academic integrity in Medical Universities,awareness of scientific integrity and acquisition channel of relevant knowledge.Results According to the survey,the awareness rate of research integrity is not high,scientific misconduct has occurred consciously or unconsciously.By analyzing the access and demands to knowledge of research integrity,we find that related seminars or lecture is the best channel to acquire academic research integrity knowledge,accounting for 61.48%.Followed by the curriculum of academic ethics and compliance aspects was the network information or communication with classmates,which accounts for 39.34%.The data also show that 95.90% of respondents thought it is necessary for medical graduate students to accept academic ethics and norms of education,68.03% of the respondents realized that cultivating students' awareness of scientific integrity in the process of scientific research gradually has great guiding significance for mentors and counselors.Conclusions The survey suggests that the education of scientific integrity in our country is very inadequate,the knowledge on good scientific integrity was fragmented.At present,most of the medical universities have not yet set up scientific integrity course,and access to the knowledge of integrity is limited for students.The scientific integrity education has become an extremely urgent issue for cultivating the sense of integrity in scientific research for researchers as soon as possible.

ABSTRACT

Objective To learn the prevalence situation of dyslipidemia among adult residents in the southern coastal area, Wenzhou,Zhejiang.Methods Adult residents were chosen by using multi -stage sampling method from 11 counties in Wenzhou in 2013.Questionnaire survey,medical examination,and biochemical detection triglycerides were conducted among the residents.Chi -square test,and multiple logistic regression analysis was used to identify the risk factors related to dyslipidemia.Results The overall prevalence of dyslipidemia was 44.99%(the standardized prevalence rate was 42. 93%),and prevalence rate in women (43.91%)was higher than that in men (45.80%),but the urban(14.26%)and rural(13.83)areas prevalence was not significant.The awareness rate of dyslipidemia was only 21.73%.The prevalence of isolated low HDL -C,high LDL -C,hypercholesterolemia,hypertriglyceridemia,mixed hyperlipidemia was 13.95%, 10.45%,19.34%,12.98%,4.69%,respectively.Multivariate logistic regression analysis showed that gender,age, nationality,coronary heart disease,dyslipidemia,family history,aquatic products,milk,pickled products edible frequency,living and working pressure,body mass index,central obesity,hypertension,diabetes were related the prevalence of dyslipidemia,but the risk factors of different lipid fractions were different.Conclusion The prevalence of dyslipidemia has been in a high level among adult residents in Wenzhou,but the awareness rate of dyslipidemia was low. Measures should be Strengthened to prevent dyslipidemia .

ABSTRACT

BACKGROUND:Partial pressure of end-tidal carbon dioxide (PETCO2) has been used to monitor the effectiveness of precordial compression (PC) and regarded as a prognostic value of outcomes in cardiopulmonary resuscitation (CPR). This study was to investigate changes of PETCO2 during CPR in rats with ventricular fibrillation (VF) versus asphyxial cardiac arrest. METHODS:Sixty-two male Sprague-Dawley (SD) rats were randomly divided into an asphyxial group (n=32) and a VF group (n=30). PETCO2 was measured during CPR from a 6-minute period of VF or asphyxial cardiac arrest. RESULTS:The initial values of PETCO2 immediately after PC in the VF group were significantly lower than those in the asphyxial group (12.8±4.87 mmHg vs. 49.2±8.13 mmHg,P=0.000). In the VF group, the values of PETCO2 after 6 minutes of PC were significantly higher in rats with return of spontaneous circulation (ROSC), compared with those in rats without ROSC (16.5±3.07 mmHg vs. 13.2±2.62 mmHg,P=0.004). In the asphyxial group, the values of PETCO2 after 2 minutes of PC in rats with ROSC were significantly higher than those in rats without ROSC (20.8±3.24 mmHg vs. 13.9±1.50 mmHg,P=0.000). Receiver operator characteristic (ROC) curves of PETCO2 showed significant sensitivity and specificity for predicting ROSC in VF versus asphyxial cardiac arrest. CONCLUSIONS:The initial values of PETCO2 immediately after CPR may be helpful in differentiating the causes of cardiac arrest. Changes of PETCO2 during CPR can predict outcomes of CPR.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the pulmonary dysfunction patterns in patients of scoliosis associated with neurofibromatosis type I (NF1) and to identify factors affecting the pulmonary function in patients with scoliosis associated with NF1.</p><p><b>METHODS</b>Preoperative pulmonary function tests (PFTs) were evaluated in 100 patients with scoliosis [NF1 group, 36 cases; idiopathic scoliosis (IS) group, 64 cases] from January 2003 to June 2009. According to location of apical vertebra and dystrophic change in patients with NF1, the parameters of pulmonary function [vital capacity (VC), forced vital capacity (FVC), forced expiratory volume in one second (FEV1), maximal mid-expiratory flow (MMEF), maximal voluntary ventilation (MVV)] were compared between NF1 group and IS group, and between the subgroups of NF1. The correlation between pulmonary function parameters and radiographic parameters of scoliosis was analyzed.</p><p><b>RESULTS</b>The VC, FVC, FEV1, MMEF, MVV in NF1 group and IS group were of no significant difference (P > 0.05). In NF1 patients, the pulmonary dysfunction was more severe in thoracic subgroup than non-thoracic subgroup (P < 0.05), while there was no difference between dystrophic scoliosis and non-dystrophic scoliosis (P > 0.05). The location of apical vertebra and the severity of scoliosis correlated significantly with the pulmonary dysfunction in NF1 group.</p><p><b>CONCLUSIONS</b>The pattern of pulmonary dysfunction in scoliosis associated with NF1 is similar with IS. Pulmonary dysfunction is more severe in thoracic scoliosis. The location of apical vertebra and the severity of scoliosis are the risk factors influencing the pulmonary dysfunction.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To prospectively evaluate the clinical effects of posterior paramedian approach in nerve root decompression and reducing muscle damage in low back surgeries.</p><p><b>METHODS</b>Study group included 30 cases treated from January 2007 to May 2008, DDD 8 cases, spondylolisthesis 6 cases, LDH 11 cases, Low back surgery failure re-operation 5 cases. Based on the comprehensive understanding of modern spine anatomy, we abandoned laminectomy in our procedure, applied a mid-waist skin incision, dissect to the paraspinal muscles where you could easily reach the facets by separating between the multifidus and longissimus, enlarge the canal by performing resection along ligamentum flavum and the inner broader of the articular process, remove enough tissue till you could expose the traversing root and the disc space, this method could achieve a limited but precise and effective decompression with not taking out all of the articular process. Once the anatomy mark of the pedicle is located (usually would be at the central area of the incision), pedicle screws placement would be precise and easy without struggling with muscle traction. The following procedures would be Spondylolisthesis reduction, discectomy and interbody fusion.</p><p><b>RESULTS</b>Post-op patients of study group all showed significant improvement of pain symptoms, VAS reduced from 7.14 + or - 1.8, pre-op to 1.39 + or - 0.72 post-op, narrowed disc space regained height, spondylolisthesis reached anatomic reduction, no complications such as pedicle screw misplacement and nerve root damage were found, the lumbar spine regained it's physiological lordosis structure. Significant difference is discovered (P < 0.001) in statistic study concerning the rate of intractable low back pain between pre-op and post-op.</p><p><b>CONCLUSIONS</b>Applying low back surgery through posterior para-median approach could directly reach the inferior/superior facets and the "soft" structures of the spinal canal, expose the exact decompression region and anatomy mark of the pedicle in the central surgical field without strong retraction on the para-spinal muscles. This approach has the advantage of lowering the surgical difficulty of implantation, reducing the risk of nerve damage and is also a minimum invasive procedure. In many cases, laminectomy is unnecessary, leaving the lamina intact could preserve the physiological anatomy of the spine.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of the extract from bergamot and boxthorn on the skin and the hair growth in mice.</p><p><b>METHOD</b>The skin on the back of mice was shaved topically and smeared with bergamot and boxthorn extract for 42 days. Then the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) and collagen in the mice were measured. The growth of hair was also observed.</p><p><b>RESULT</b>Compared with control group, the extract from bergamot and boxthorn obviously increased the activity of superoxide dismutase (P < 0.05) and the content of collagen (P < 0.001), and decreased the content of malondialdehyde (P < 0.05) in the skin of mice. It also significantly promoted the growth of hair (P < 0.001).</p><p><b>CONCLUSION</b>The extract from bergamot and boxthorn plays an active role in skin and the promotion of hair growth.</p>
